台盼蓝,重氮染料是一种重要的染色剂,可以将死组织或细胞染成蓝色。具有完整细胞膜的活细胞或组织不着色。由于细胞在通过膜的化合物中具有很高的选择性,因此在活细胞中,台盼蓝不被吸收; 然而,它穿过死细胞中的膜。因此,死细胞在显微镜下显示为独特的蓝色。台盼蓝通常用于显微镜检查(用于细胞计数)和实验室小鼠用于评估组织活力。该方法不能区分坏死细胞和凋亡细胞。观察真菌和Stramenopiles的排泄物也很有用。

生物素修饰的脱氧尿苷5′-三磷酸广泛用于各种非放射性DNA标记反应,包括切口平移,随机引物标记,cDNA标记和3’末端标记。已显示生物素化的探针以与非生物素化探针相同的速率和程度与同源核酸杂交。杂交的生物素化DNA探针可以通过抗生物素蛋白和链霉抗生物素蛋白检测。生物素-11-dUTP可以通过切口平移,随机引发,3′-末端标记或在PCR过程中酶促地掺入DNA中。数字’11’是dUTP和生物素之间的接头主链中的碳原子数。接头越长,生物素与抗生物素蛋白的相互作用就越有效。另一方面,接头越短,dUTP更有效地掺入DNA。建议连接子’11’的长度对于大多数应用是最佳的。 Bio-11-dUTP用于在各种杂交应用中生产生物素化的DNA探针,包括Southern印迹,Northern印迹,斑点印迹,固定细胞和组织。
