产品说明书

样品实验方案

工作溶液配制

Gelite Safe 工作溶液

在7.5-8.5的pH范围内使用(例如,TAE、TBE或TE pH 8.2)缓冲液稀释10,000X储备试剂来制备1X Gelite Safe工作溶液。
注意:在水中配制的染色溶液比在缓冲液中配制的溶液不稳定,必须在24小时内使用。

 

后染色方案
1.根据您的标准方案运行凝胶。
2.将凝胶放在合适的聚丙烯容器中。 轻轻加入足量的1X染色溶液以浸没凝胶。
注意:请勿使用玻璃容器,因为它会吸收染色溶液中的许多染料。
3.在室温下避光轻轻搅拌凝胶约30至60分钟。
4.使用300 nm / 254 nm紫外透射仪或使用长距离绿色滤光片(例如SYBR®滤光片,GelStar®滤光片,GelGreen®滤光片或GelRed®滤光片)的凝胶扫描仪对凝胶成像。

 

预染色方案
1.使用标准方案准备琼脂糖凝胶溶液。
2.在倒入凝胶之前,将10,000X Gelite Safe储备试剂以1:10,000的比例稀释到凝胶溶液中并彻底混合。
3.根据您的标准方案运行凝胶。
4.使用300 nm / 254 nm紫外透射仪或使用长距离绿色滤光片(例如SYBR®滤光片,GelStar®滤光片,GelGreen®滤光片或GelRed®滤光片)的凝胶扫描仪对凝胶成像。

与常用的DNP-X酸(#2020)相比,DNP-PEG4酸具有更好的水溶性。它是可被抗DNP抗体识别的探针的胺反应性构建基块。它也可以用作与Trp或Tyr配对的出色的胺反应性FRET淬灭剂。在EDC和DCC存在下,其末端羧基容易与伯胺反应形成稳定的酰胺键。亲水性PEG接头增加了化合物在水性介质中的水溶性。

溶液配制

储备溶液配制 

iFluor 594 小麦胚芽凝集素 (WGA) 缀合物储备液 (200X)

将 500 µL ddH2O 添加到粉末小瓶中,制成 2 mg/mL 的储备溶液。 注意:重新配制的缀合物溶液可在 2-8 °C 下短期储存,或在 -20 °C 下长期储存。

工作溶液配制 

iFluor 594-小麦胚芽凝集素 (WGA) 缀合物​​工作溶液 (1X)

将 5 μL 200X WGA 储备溶液添加到 1 mL HHBS 缓冲液中。
注意:不同细胞系的优化染色浓度可能不同。 活细胞的推荐起始浓度为 5-10 µg/mL。

 

操作步骤

1.活细胞染色
1.1 用 HHBS 缓冲液清洗细胞两次。
1.2 添加 100 µL iFluor 594-WGA 工作溶液。
1.3 将细胞与 WGA 工作溶液在 37°C 下孵育 10-30 分钟。
1.4 用 HHBS 缓冲液清洗细胞两次。
1.5 使用 Cy3/TRITC 滤光片组在荧光显微镜上成像细胞。

 

2.固定细胞染色
2.1 在PBS中使用4%的甲醛固定细胞。
注意:对于固定细胞膜染色,建议在没有透化步骤的情况下进行染色。固定后透化步骤会导致细胞内区室染色,如高尔基体和内质网 (ER) 结构染色。
2.2 添加 100 µL iFluor 594-WGA 工作溶液。
2.3 在室温下用 WGA 工作溶液孵育细胞 10-30 分钟。
2.4 用 HHBS 缓冲液清洗细胞两次。
2.5 使用 Cy3/TRITC 滤光片组在荧光显微镜上成像细胞。
