产品介绍
钙黄绿素深红是美国AAT Bioquest生产的钙指示剂,钙黄绿素AM是最流行的荧光探针之一,用于标记和监测活细胞的细胞功能。但是,钙黄绿素AM的单色使得无法在多色应用中使用这种有价值的试剂。例如,由于与GFP相同的颜色,不可能在GFP转染的细胞中使用钙黄绿素AM。为了解决钙黄绿素AM的颜色限制,我们开发了钙黄绿橙等一系列产品。这些新的Calcein AM类似物与Calcein AM结合使用,可以对活细胞进行多色标记和功能分析。钙黄绿素深红TM是钙黄绿素深红 乙酸酯在细胞中的酯酶水解产物。钙黄绿素Deep Red 染料可以使用通用的Cy5滤波片进行检测。它用作Calcein Deep Red 乙酸酯的荧光参考标准。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的钙黄绿素深红。

产品说明书
样品实验方案
简要概述
1.在生长培养基中制备细胞
2.添加Amplite ROS绿色工作溶液(96孔板100μL/孔或384孔板25μL/孔)
3.将细胞在37℃下染色60分钟
4.用测试化合物处理细胞以诱导ROS
5.在Ex / Em = 490/525 nm(截止= 515 nm)或带有FITC滤光片组的荧光显微镜下监测荧光增加(底部读取模式)
溶液配制
1.储备溶液配制
除非另有说明,否则所有未使用的储备溶液应分成一次性等分试样,并在制备后储存在-20°C。 避免反复冻融循环。
1. 1Amplite ROS Green原液(500X):
将40μLDMSO(组分C)加入到Amplite ROS Green(组分A)的小瓶中并充分混合以制备500X Amplite ROS Green原液。 避光。 注意:20μL的500X Amplite ROS Green原液足以容纳1个平板。
2.制备工作溶液
将20μL500XAmplite ROS Green储备溶液加入10 mL分析缓冲液(组分B)中,充分混合,制成Amplite ROS Green工作溶液。 注意:此Amplite ROS Green工作溶液在室温下稳定至少2小时。有关细胞样品制备的指南,请点击查看。
操作步骤
1.将100μL/孔(96孔板)或25μL/孔(384孔板)的Amplite ROS Green工作溶液加入细胞板中。
2.将细胞在5%CO2,37℃培养箱中孵育1小时。
3.用所需缓冲液(如PBS或HHBS)中的20μL11X测试化合物(96孔板)或10μL6X测试化合物(384孔板)处理细胞。 对于对照孔(未处理的细胞),加入相应量的化合物缓冲液。
4.为了诱导ROS,将细胞板在室温下或在5%CO2,37℃培养箱中孵育至少15分钟或所需的一段时间(对于用1mM H2O2处理的Hela细胞30分钟)。
5.使用荧光酶标仪(底部读取模式)在Ex / Em = 490 / 525nm(截止= 515nm)监测荧光增加,或使用具有FITC滤光器组的荧光显微镜观察细胞。
